I just bought a 65 dodge D-100 from a dealer who had it in the back lot for about 300$ and it runs great after a full tune up. But my question is this:
Can i swap out the 3sd manual transmission for a 4 spd manual transmission out of my 64 dodge d100 that has a has a 2 barrel v8 (threw a rod). What do i need to do it?

I think it should be a straight swap, bellhousing/clutch/etc should swap right over to the new tranny, as long as the spline count is the same. It looks like both the bell houseing from the 65 slant six and the 64 with the v8 are nearly the same, and on the back of the bell houseing of the /6 it has the 4 bolt holes that should line up with the 4spd transmission. Is the 4 spd going to be any longer, enough that i would need to shorten the drive shaft?

The 64 with the 4spd is a long bed, and the 65 with the 3spd is a short bed. Either way i would still have an extra drive shaft that i can have shortend.
