Has anyone done a comparison of Edelbrock carb mounting on a Offie manifold? Which worked better, for drag racing, primaries forward, or primaries toward fender?

No back-back comparison for me, but I have run a few different carbs both ways, incl Edel 4bbl. I would guess 0.1 sec or less difference.

I have no data, but my instinct says that the orientation would affect part-throttle distribution (street) more than WOT (drags).

The only other factor might be slosh out the vents on a hard launch if the carb is sideways, but again I have no data.

I just got an Offie Manifold, and 500 cfm Eddy. It would be easier to install nitrous nozzles in the runners, if the primaries are towards the fender. My hommade nitrous fogger manifold, just had a 2bbl Holley.

That is the way I have run those carbs unless I am doing hard cornering. One direction of hard cornering (left, IIRC) makes the car die...

When I was racing dirt hobby stock cars. We had to run a Holley 1850 with the side hung floats. One of my buddies mounted his backward with a bell crank throttle rod so the fuel level would get higher in the corners and not lower. It got some strange looks but no one ever protested it.
I just set my float level a little to the high side and didn't have any problems though.

Mike and Blaine did some testing years ago and found no difference on a drag car.
