If you remove the bottom leaf from these 5-leaf springs does that immediately lower the ride height, or does it just change the spring's reaction to bumps or carrying a heavy load?

The car weighs 2700-2750 pounds and the springs are JC Whitney 5-leaf
How much should it lower the rear?

Yes and yes - you're taking stiffness out of the spring. Not sure how much drop you'll get.
