Not always, I'd gotten a 7 1/4 to spin both at the same time and I new it was an open diff. If your planning on making 300hp you should step up to an 8 1/4 at least.

It will not spin one tire, if it is working corectly.
It can handle 250 hp IF it is a sure grip, and if it has been put together properly, and not worn out. Only with an auto, not a stick. It will not live with shock loading.

Try to find an 8 1/4 rear, they are pretty easy to find. A good 8 1/4 can withstand a hot slant no problem. I have seen posi 8 1/4 survive behind 383's before.

Just read the posts, on this forum, and ask the right questions. It's actually fairly easy, with power adders, but can be done, normally aspired, also. My 170 makes in the upper 200hp range, on N2O.

Id agree with Charlie on his comment here. You can get an easy 300hp out of a slant by concentrating on the right combination of parts- Like any engine. Id focus on a very nice cylinder head- porting and bigger valves.]
The cylinder head is the key and is where you gonna make your power.
:D

As far as the 7.25 goes I doubt it could handle 300hp unless it was a sure-grip. I seen Charrlie' thrashin that cuda in the 2002 slant 6 shootout here in Wilkesboro and he was laying down in the 8.20's (IIRC)with his 7.25 sure grip rear on the slicks. They are fairly light rears but good luck finding the sure-grip 7.25- Scarce to say the least. I've seen 2 - Including
the one Im building for my duster! An 8.25 sure grip would be a good idea
though. Building one of those for the '66 cuda bracket car...in progress and will be for quite a while. Good luck with your project!
